Cloning and Functional Analysis of NtMYB9 in ‘Jinzhanyintai’ of Narcissus tazetta var. chinensis

Narcissus tazetta var. chinensis M.Roem. (Chinese Narcissus) is a traditional and famous flower in China, its corona shows transition from green to yellow with the opening of flowers, which great ornamental value. To investigate molecular mechanism color-forming Chinese Narcissus, this study, MYB gene was screened transcriptome by bioinformatics analysis methods, named NtMYB9. The expression level NtMYB9 significantly higher than tepal, filament, ovary leaf, reached highest at fourth period complete coloration corona. full-length sequence coding region cloned using cDNA as template. Sequence revealed that protein contained R2 R3 domains, phylogenetic tree showed most closely related flavonol activators. Subcellular localization localized nucleus. overexpression into tobacco leaves seven enzyme genes flavonoid biosynthesis pathway were up-regulated. transient transformation Petunia × hybrida (Petunia) Phalaenopsis aphrodite (Phalaenopsis) could turn flowers yellow. These results suggest involved positive regulation may promote synthesis flavonols. In conclusion, these findings provide valuable resource for further studies on regulatory pathway, they are also beneficial breeding Narcissus.
